I think it takes 3-4 hours, depending on speed and traffic. However, this trip is probably the single least appealing to do by bike in Cambodia. National road 4 has crazy traffic, including heavy trucks and speeding vacationers. If you do want to ride to Sihanoukville, consider the detour through Kampot on national road 3, or better yet, the road through Kep.
